Alpha-fetoprotein (AFP), a glycoprotein belonging to the albumin family, is synthesized by fetal liver cells and yolk sac. Alpha-fetoprotein has a high concentration in the fetal blood circulation, which decreases after birth, and is basically replaced by albumin in 2 ~ 3 months after birth. It is difficult to detect alpha-fetoprotein in blood, so the content in adult serum is very low. Alpha-fetoprotein has many important physiological functions, including transport function, bidirectional regulation as a growth regulator, immunosuppression, T-lymphocyte induction of apoptosis and so on. Alpha-fetoprotein is closely related to the occurrence and development of liver cancer and a variety of tumors. It can show high concentration in a variety of tumors and can be used as a positive detection index for a variety of tumors. Clinically, it is mainly used as serum marker of primary liver cancer for diagnosis and therapeutic monitoring of primary liver cancer.

Singclean Alpha-fetoprotein (AFP) assay kit (ELISA) is based on an enzyme-linked immunosorbent sandwich assay (ELISA). Standards, samples and anti-AFP-HRP conjugate are added to the wells coated with Anti-AFP monoclonal antibody. AFP in the serum binds to antibody coated on the well, and then the anti-AFP-HRP antibody binds to AFP. Unbound proteins are washed off. Upon the addition of the tetramethyl-benzidine (TMB) substrate, the optical density (OD) is proportional to the concentration of AFP in the samples. A standard curve is prepared by plotting option density (OD) to the concentration of the AFP. Then read-off unknown sample values on the standard curve.
